Capsaicin determination from pain patch for the calculation of Scoville heat units by gas chromatography-mass spectrometry

Background and Aims: A sensitive, accurate precise method has been developed for the determination of Capsaicin from pain patches by Gas chromatography Mass Spectrometry (GC-MS). irritant effects in high concentrations, so these can be minimized knowing amount present efficacy safety patches. Methods: was extracted using liquid-liquid extraction The Chromatographic separation performed 5% diphenyl 95% dimethylpolysiloxane column with a purity 2 mL/min flow rate helium gas. made gradient oven temperature program. started at 250°C increased to 275°C 10°C.min-1 ramp held 2.5 min. injection port adjusted 300°C split mode used. analysis carried out 5:1. MS ionization potential determined 70 eV. Results: calibration curve found linear range 5 - 50 μg/mL. limits detection quantification capsaicin 3.46 μg/mL μg/mL, respectively. validated successfully applied patch analysis. Conclusion: This is simple, reproducible, used safely routine without derivatization. study how calculate Scoville Heat Units (SHU) that contain Capsaicin. patch, its effects, appear low when evaluated SHU.
